Ecstasy tablets are as a psychoactive drugs that use by young adult a lot. It effects on many of neurotransmiters specially serotonin . It has a lot of bad effects on differet part of body like braine and heart. In this research the effects of ecstasy on pituitary- gonad axis and spermatogenesis in male rat were studied. For experimenta l and control groups 50 adult male rats from NMRI race were divided in to 5 groupsof 10 . Because of effective substances (MDMA) in this tablets had not determined , at first we scaled them then attenuated them with normal saline in 3 levels of concentrations ( 2.5, 5, 10 mg/kg ) were prepared. The experimental groups (C,D,E)were given oral doses of it by special feeder for a period of 28 days and sham group (B) received saline and control group did not receive any thing . The blood samples were taken after 28th day and the concentration of LH, FSH, Testostrone were measured by RIA. In addition at the 28th day the testes were separated and histological changes was studied among experimental and control groups . The results were evaluated by using ANOVA,t-test,post hoc test ,Duncan test . but concentration of LH did not show significant difference in compared with the control group .Ecstasy has a high affinity to H1 histamine receptor basal level of LH did not change because of this mechanism .Ecstasy increases to release of serotonin via this mechanism maybe FSH secretion increases in low dose In high dose of experimental groups shows a significant decrease in concentration of FSH hormone in comparison with control group because of activation of feedback mechanisms . Ecstasy causes an increased in concentration of testosterone via increasing the secretion of serotonin or via increase in LH receptor or likely it can stimulate prolactin secretion that can increases the testosterone secretion . Ecstasy causes atrophy in testis . Perhaps hyperthermy induced ecstasy has a direct effects on spermatogenesis and it can degenerate spermatogenesis. Suggested that ecstasy effect on thermoregulation region in hypothalamous and also it can induce hyperactivation of Hypothalamus-pituitary - thyroid axis and sympathethic system that all of them induce hyperthermia.
